dc.descriptionThe present investigation was carried out in Cotopaxi Province, in Salcedo Canton´s neighborhoods, the objective of the study was to sequence the internal transcribed spacer region (ITS-2) of the ribosomal DNA (rDNA) of Toxocara canis. A total of 106 canine stool samples were collected from adults and puppies of both sexes. The population ranged in age from one month until nine years. The samples were taken from 3 to 5 grams of feces, the samples were taken in the laboratory of the Technical University of Cotopaxi in the Biology Laboratory, under the Sheather flotation technique. Sixteen canines were positive, showing the presence of Toxocara canis eggs (16.96 %) of prevalence according to the samples taken and analyzed by microscopy. To obtain the parasite, the population under study was orally dewormed, taking into account the nutritional and living conditions of the pet. The adult parasite was collected from 24 canines, indicating a 25.44 % presence of the nematode, based on the macroscopic morphology. In 8 canines from one month to six months old, deworming was performed without coprological examinations where parasites were expelled. The 20 samples isolated from canines were identified as Toxocara canis when compared using BLAST with sequences previously deposited in GenBank with 100% identity for T. canis. For the sequencing of the adult male parasite, it was proceeded to dissect following the process of preservation, the integrity of the biological sample, which guarantees the amplification, purification and sequencing of DNA, avoiding the dissection of reproductive organs and causing interference at the time of amplification of DNA fragments. The tests performed indicate high reliability in the diagnosis of molecular tools.es_ES
dc.description.abstractLa presente investigación se efectuó en la Provincia de Cotopaxi, en los barrios del Cantón Salcedo, el objetivo de estudio fue realizar la secuenciación de la región del espaciador transcrito interno (ITS-2) del ADN ribosómico (ADNr) del Toxocara canis. Se recolectaron 106 deposiciones de caninos, entre adultos y cachorros de ambos sexos. La población tiene un rango de edad entre un mes a nueve años. Se tomaron de 3 a 5 gramos de heces, las muestras fueron realizadas en el laboratorio de la Universidad Técnica de Cotopaxi en el Laboratorio de Biología, bajo la técnica de flotación de Sheather. Resultaron positivos 16 caninos, mostrando la presencia de huevos de Toxocara canis, (16.96 %) de prevalencia según las muestras tomadas y analizadas mediante microscopio. Para la obtención del parasito se desparasito vía oral a la población en estudio, tomando en cuenta las condiciones nutricionales y condiciones que habita la mascota, se recolecto de 24 caninos el parasito adulto, indica un 25.44 % la presencia del nematodo, basado en la morfología macroscópica. En 8 caninos de un mes a seis meses se realizó la desparasitación sin exámenes coprológicos donde hubo expulsión de parásitos. Las 20 muestras aisladas de caninos se identificaron como Toxocara canis cuando se compararon usando el BLAST con secuencias previamente depositadas en el GenBank con un 100% de identidad para T. canis. Para la Secuenciación del parasito macho adulto se procedió a diseccionar siguiendo el proceso de preservación, la integridad de la muestra biológica, que garantiza la amplificación, purificación, secuenciación del ADN, evitando que se diseccione órganos reproductores y causen interferencia al momento de realizar la amplificación de fragmentos del ADN. Los exámenes realizados indican la alta confiabilidad en el diagnóstico de herramientas moleculares.es_ES
